The study was intended to find the pharmacodynamic and pharmacokinetic drug interaction of class Ib antiarrhythmic drug mexiletine with tolbutamide in rabbits following single and multiple dosage treatment. The interaction studies were carried out in rabbits. Therapeutic dose (TD) dose of mexiletine, TD of tolbutamide were administered to same group of animals and blood glucose was estimated by GOD/POD method. In case of combination TD of mexiletine and TD of tolbutamide were administered and blood glucose as well as tolbutamide were estimated for pharmacokinetic data. Tolbutamide was estimated by DNFB method and insulin was estimated by radioimmunoassay. The studies conducted in rabbits both in single and multiple doses indicated enhancement of tolbutamide effect. The drug mexiletine alters the pharmacokinetics of tolbutamide and increases the insulin levels. The studies of interaction between mexiletine and tolbutamide indicate enhanced the absorption of tolbutamide in rabbits. Mexiletine increased insulin secretion in rabbits. The mexiletine appears to produce interaction with tolbutamide by pharmacodynamic and pharmacokinetic mechanisms.
